What the research says about Siberian Ginseng, Rhodiola, Ashwagandha and Linden
Educational summaries of published research on this formula's herbs generally; not product-specific claims. Orange Naturals Stress Tincture's own authorized use under NPN 80036436 is that it is used in Herbal Medicine to help improve mental and/or physical performance after periods of mental and/or physical exertion, and as a tonic to help relieve general debility and/or to aid during convalescence.
A review of 46 USSR-era human trials found Siberian Ginseng benefited endurance, cognitive function and infection resistance
This narrative review recovered and translated 46 Soviet-era human clinical trials (1962-1986) on Eleutherococcus senticosus, most in healthy volunteers at a typical dose of about 2 mL extract/day, examining physical and mental stamina, cold/influenza incidence and cognitive effects. The authors concluded ES appears to benefit cognitive function and physical/mental endurance, with promising results for respiratory-infection prophylaxis, supporting its traditional adaptogen and tonic use.
Narrative Review (46 human trials)A randomized trial found Siberian Ginseng modestly and transiently improved quality of life in elderly adults
20 elderly hypertensive volunteers were randomized to Eleutherococcus senticosus extract (300 mg/day) or placebo for 8 weeks, assessed with the SF-36v2 quality-of-life questionnaire. After 4 weeks, the treatment group showed a significantly higher social-functioning score than placebo, though this difference did not persist to the 8-week mark - an honest, transient finding reported as-is, with no adverse events observed in either group.
Human RCT (n=20)A pharmacological review catalogued Siberian Ginseng's traditional 'invigorating' use and its anti-stress, anti-fatigue research
This comprehensive review of Acanthopanax senticosus (Siberian Ginseng) botany, chemistry and pharmacology summarized its traditional Chinese Medicine use to 'invigorate qi, strengthen the spleen and nourish kidney,' and catalogued preclinical and clinical evidence for anti-stress, anti-fatigue, anti-inflammatory and immune-modulating activity, positioning it alongside Panax ginseng as a widely used adaptogen.
ReviewA 144-person trial found adding Siberian Ginseng to stress-management training gave no additional benefit over the training alone
144 participants with stress-related asthenia and reduced working capacity were randomized to 120 mg/day Eleutherococcus senticosus extract, a 2-day professional stress-management training program, or both combined. Nearly all outcome measures (cognitive performance, stress, fatigue, mood, sleep, cortisol awakening response) improved significantly over time regardless of group, with no meaningful advantage from adding the herbal extract to the training - an honest null finding for this specific comparison, though all treatments were well tolerated.
Human RCT (n=144)A different commercial combination formula containing Siberian Ginseng significantly improved burnout symptoms over 12 weeks
87 workers with professional burnout syndrome were randomized to a different commercial supplement (combining casozepine, taurine, Eleutherococcus senticosus and extramel) or placebo for 12 weeks. The active-formula group showed significant improvement across all five measured burnout/depression scores, consistently outperforming placebo - a real result for that specific combination product, not this product's own formula, included here as broader real-world evidence for Siberian Ginseng's role in multi-ingredient stress formulas.
Human RCT (n=87)A pharmacology review found adaptogens including Siberian Ginseng act on the HPA axis to increase tolerance to mental exhaustion
This review assessed the level of clinical evidence for adaptogens in fatigue, concluding good scientific evidence exists for Schisandra chinensis and Eleutherococcus senticosus increasing endurance and mental performance in patients with mild fatigue and weakness, via mechanisms linked to the hypothalamic-pituitary-adrenal (HPA) axis, molecular chaperone Hsp70, and regulation of circulating cortisol.
ReviewA 60-person trial found Rhodiola rosea extract reduced burnout-related fatigue and cortisol response to awakening stress
60 individuals meeting diagnostic criteria for stress-related fatigue syndrome were randomized to a standardized Rhodiola rosea extract (SHR-5, 576 mg/day) or placebo for 28 days. The Rhodiola group showed significantly greater improvement on Pines' burnout scale and several attention-test indices, along with a significant change in saliva cortisol response to awakening stress compared to placebo, with no serious side effects attributed to the extract.
Human RCT (n=60)A crossover trial found repeated low-dose Rhodiola rosea improved mental performance in physicians during night duty
56 young, healthy physicians underwent a double-blind crossover trial of a standardized Rhodiola rosea extract (SHR-5) versus placebo during night-duty shifts, testing associative thinking, short-term memory, calculation and audio-visual perception speed as a composite Fatigue Index. A statistically significant improvement in these cognitive-fatigue tests was observed during the treatment period, with no side effects reported.
Human RCT, Crossover (n=56)A pilot trial found Rhodiola rosea improved physical fitness and mental fatigue in students during exam stress
Foreign students undergoing a stressful examination period took a standardized Rhodiola rosea extract (SHR-5) or placebo for 20 days in this double-blind, randomized pilot trial. The Rhodiola group showed the most significant improvement in physical fitness, mental fatigue and neuro-motor test scores, along with better self-assessed general well-being, though the authors noted the dose level was probably suboptimal.
Human RCT, PilotA 161-cadet trial found two doses of Rhodiola rosea produced a significant antifatigue effect versus placebo
161 military cadets were randomized to one of two doses of a standardized Rhodiola rosea extract (SHR-5) or placebo in this double-blind trial measuring an antifatigue index (AFI) alongside pulse rate and blood pressure. Both verum groups showed a significantly higher AFI than placebo (p < 0.001), with no significant difference between the two active doses, though a possible trend favored the lower dose on psychometric tests.
Human RCT (n=161)An open-label trial found Rhodiola rosea extract improved symptoms in people with prolonged or chronic fatigue
100 subjects with prolonged or chronic fatigue symptoms took 400 mg/day of a Rhodiola rosea extract for 8 weeks in this open-label multicenter trial. The greatest improvement was seen after just 1 week, with fatigue symptoms continuing to decline through a statistically significant improvement by week 8; most adverse events were mild and unrelated to the study drug.
Human Trial, Open-Label (n=100)A head-to-head trial found Rhodiola rosea produced a smaller antidepressant effect than sertraline, but with fewer side effects
57 adults with mild-to-moderate major depressive disorder were randomized to 12 weeks of a standardized Rhodiola rosea extract, sertraline, or placebo. Sertraline produced a numerically larger reduction in depression scores than Rhodiola, but significantly more participants on sertraline reported adverse events (63.2%) than on Rhodiola (30.0%) or placebo (16.7%) - an honest finding that Rhodiola was less effective than a standard antidepressant, but may offer a more favorable risk-to-benefit ratio for milder cases.
Human RCT, Phase II (n=57)A 64-person trial found high-concentration Ashwagandha root extract significantly reduced stress scores and serum cortisol
64 adults with chronic stress took 300 mg of a high-concentration full-spectrum Ashwagandha root extract or placebo twice daily for 60 days in this randomized, double-blind trial. The Ashwagandha group showed a significant reduction (p<0.0001) on all stress-assessment scales and a significant drop in serum cortisol (p=0.0006) compared to placebo, with only mild adverse effects comparable between groups.
Human RCT (n=64)A 60-day trial found Ashwagandha reduced anxiety and morning cortisol, with a near-significant reduction in overall stress scores
60 stressed, healthy adults took 240 mg of a standardized Ashwagandha extract (Shoden) or placebo once daily for 60 days. Ashwagandha produced a statistically significant reduction in the Hamilton Anxiety Rating Scale (p=0.040) and a near-significant reduction in the Depression, Anxiety and Stress Scale (p=0.096) versus placebo, plus significantly greater reductions in morning cortisol and DHEA-S - an honestly reported partial result on the broader stress-scale measure, with no adverse events.
Human RCT (n=60)A meta-analysis of 9 trials found Ashwagandha significantly reduced perceived stress, anxiety scores and cortisol versus placebo
Pooling 9 randomized controlled trials (558 patients), this systematic review and meta-analysis found Ashwagandha formulations significantly improved Perceived Stress Scale scores, Hamilton Anxiety Scale scores and serum cortisol levels compared to placebo, with only mild-to-moderate adverse events reported in 4 of the included studies and no major safety concerns identified.
Meta-Analysis (9 RCTs, n=558)A Bayesian meta-analysis found Ashwagandha supplementation was more effective than placebo for physical performance measures
This systematic review and Bayesian meta-analysis of 13 clinical trials through 2020 evaluated Ashwagandha's effect on strength/power, cardiorespiratory fitness and fatigue/recovery in healthy adults. The Bayesian hierarchical models found Ashwagandha supplementation was more efficacious than placebo across the analyzed physical-performance variables in both men and women, though the authors called for more comparable studies in athletic populations to derive a more precise effect estimate.
Systematic Review, Bayesian Meta-Analysis (13 trials)A three-level meta-analysis found Ashwagandha improved exercise performance on average, but benefits were not consistent across all contexts
Pooling 13 randomized trials (599 participants, 79 effect sizes), this meta-analysis found Ashwagandha improved overall exercise performance versus placebo (Hedges' g=0.47), most consistently for aerobic endurance, with 500-600 mg/day the most evidence-supported dose. However, the 95% prediction interval for the overall effect crossed zero, meaning the benefit is not uniform across every context - an honest statistical caveat alongside the positive average finding.
Systematic Review, 3-Level Meta-Analysis (13 RCTs, n=599)A meta-analysis of 5 trials found Ashwagandha significantly reduced anxiety scores and improved several sleep measures
Pooling 5 randomized controlled trials (254 patients), this meta-analysis found Withania somnifera significantly reduced Hamilton Anxiety Scale scores and improved sleep-onset latency, total sleep time and sleep efficiency versus placebo, though not wake-after-sleep-onset or total time in bed - the authors called for larger trials to confirm these promising but still-limited findings.
Meta-Analysis (5 RCTs, n=254)A 2026 review detailed the molecular mechanisms by which adaptogens including Siberian Ginseng, Rhodiola and Ashwagandha regulate the HPA axis
This review examined the molecular mechanisms of seven key plant adaptogens, finding they regulate the hypothalamic-pituitary-adrenal (HPA) axis, induce protective heat-shock proteins (Hsp70), and modulate stress-signaling pathways - with clinical studies confirming reductions in cortisol levels of 14-30%, decreased fatigue and improved cognitive function without adverse effects across the reviewed trials.
ReviewA clinical review of the most-studied adaptogens found consistent stress-protective and stimulative effects on the neuroendocrine system
Focusing on the three most extensively studied adaptogens - Rhodiola rosea, Eleutherococcus senticosus and Schisandra chinensis - this review discussed clinical evidence for stress-protective and mildly stimulating effects, along with the molecular mechanisms by which these herbs act on metabolic and neuroendocrine regulatory processes.
ReviewA historical review traced 50+ years of adaptogen use in sports and medicine, citing benefits for chronic fatigue, cognition and immune protection
This historical review described how plant adaptogens including Eleutherococcus, Rhodiola and Schisandra were first used in professional sports roughly 50 years ago for their potential to increase resistance to stress and improve physical endurance, and summarized meta-analysis data showing benefits for chronic fatigue, cognitive impairment and immune protection - while noting that human clinical trials overall remain limited relative to historical use.
Historical Review, Meta-AnalysisA study of a related Tilia species found a purified flavonoid fraction had dose-dependent anxiolytic activity in mice
This study isolated a flavonoid-rich fraction (tiliroside, quercetin, quercitrin, kaempferol and their glycosides) from Tilia americana var. mexicana - a DIFFERENT Tilia species from this product's own Tilia cordata, traditionally used in Mexican folk medicine for nervous disorders and insomnia. In mice, the fraction produced dose-dependent anxiolytic activity in the elevated plus-maze test starting at 25 mg/kg, without affecting motor activity, supporting the plant genus's traditional nervine reputation.
Preclinical Study (mice)A follow-up study found the same Tilia flavonoid fraction interacts with serotonin receptors to produce its anxiolytic effect
Extending the prior finding in a different Tilia species (Tilia americana var. mexicana, not this product's own Tilia cordata), this study found the standardized flavonoid fraction's anxiolytic effect in mice was enhanced by a serotonin 5-HT2A agonist and blocked by a 5-HT2A antagonist, and was reduced by blocking 5-HT1A receptors - suggesting the traditional Tilia genus's calming reputation may work partly through the serotonergic system.
Preclinical Study (mice)A rat study found a linden-flower infusion could not meaningfully counter ethanol-induced oxidative liver stress - an honest negative finding
Testing an infusion of linden flowers (Tilia platyphyllos - a different Tilia species from this product's own Tilia cordata, and testing a liver-protection claim this product's own NPN does not carry) against ethanol-induced oxidative stress in rats, this study found the infusion did not restore antioxidant defense markers or reduce lipid oxidation to control levels, concluding the linden infusion tested could not be considered as important as diet-derived antioxidants for preventing this specific type of oxidative damage - an honest negative preclinical result included here for research transparency.
