What the research says about magnesium and bis-glycinate chelation

Educational summaries of published research on magnesium and its bis-glycinate (glycinate) chelate form generally - shared for education, not as claims about this finished product. Pediatric-specific magnesium research is more limited than adult research for some topics (sleep, general muscle-function mechanism); those groups draw partly on the wider adult literature, noted explicitly where relevant. Orange Naturals Kids Magnesium Chewables' own authorized use under NPN 80100801 is that it is a factor in the maintenance of good health, helping in energy metabolism and tissue formation, bone development, and normal muscle function.

Magnesium Bis-Glycinate Absorption & Chelation Mechanism (This Formula's Own Active)

In patients with impaired absorption, magnesium diglycinate was better absorbed than magnesium oxide

A randomized, double-blind crossover trial in 12 adults who had undergone ileal resection compared magnesium diglycinate (a bis-glycinate chelate) with magnesium oxide. In the four patients with the most impaired magnesium absorption, the chelate was absorbed significantly more (23.5% vs 11.8%) and was better tolerated overall, supporting the idea that part of the dose is absorbed intact via a dipeptide transport pathway.

Magnesium Bis-Glycinate Absorption & Chelation Mechanism (This Formula's Own Active)

A review describes how binding magnesium to an amino acid, as in bis-glycinate, can enhance mineral absorption

Reviewing metal-binding peptides, the authors describe how binding a mineral such as magnesium to an amino acid or short peptide can influence the resulting complex in ways that improve absorption and bioavailability relative to unbound, inorganic mineral forms - the chelation principle behind magnesium bis-glycinate.

Magnesium Bis-Glycinate Absorption & Chelation Mechanism (This Formula's Own Active)

An animal dose-ranging study found magnesium glycinate shows a distinct tissue-absorption pattern from other organic magnesium forms

Comparing organic-acid-bound magnesium (citrate, malate) with amino-acid-bound forms (acetyl taurate, glycinate) at three different doses in mice, magnesium citrate increased muscle and brain magnesium in a dose-independent way, while dividing high doses of the amino-acid-bound forms into two smaller doses did not meaningfully improve tissue magnesium levels.

A safety review found most studies of oral magnesium supplements showed no significant increase in diarrhea versus placebo

Reviewing 10 studies (5 meta-analyses, 5 randomized trials) that tracked diarrhea as an adverse event from single-ingredient magnesium supplements at doses from 128 to 1,200 mg/day, 7 found no significant difference in diarrhea between magnesium and control groups, and a search of the FDA's adverse-event database found only 40 attributable gastrointestinal cases tied to single-ingredient magnesium products.

Magnesium & Sleep (Adult Research - Pediatric-Specific Data Still Limited)

An RCT found magnesium bisglycinate - the same chelate form used in this formula - modestly improved insomnia severity in adults with poor sleep

In a randomized, double-blind, placebo-controlled trial of 155 adults aged 18-65 with self-reported poor sleep, 250 mg of elemental magnesium as magnesium bisglycinate produced a small but statistically significant improvement in Insomnia Severity Index scores over 4 weeks compared with placebo, with the largest benefit suggested in those with lower baseline dietary magnesium intake. This trial was conducted in adults; direct pediatric research on magnesium bis-glycinate and sleep specifically is still limited.

Magnesium & Sleep (Adult Research - Pediatric-Specific Data Still Limited)

A trial in adults with diabetes found magnesium supplementation reduced insomnia severity and shifted sleep hormones

In 290 adults with diabetes randomized to placebo, magnesium, potassium, or both, magnesium supplementation significantly reduced Insomnia Severity Index scores and shifted serum melatonin and cortisol levels compared with placebo over two months. This trial was conducted in adults with diabetes, not children.

An RCT in children with ADHD found vitamin D plus magnesium supplementation improved several mental-health measures

In a randomized, double-blind, placebo-controlled trial of 66 children with ADHD, 8 weeks of vitamin D (50,000 IU/week) plus magnesium (6 mg/kg/day) produced significant reductions in parent-reported emotional problems, conduct problems, peer problems and total difficulties compared with placebo.

A prospective cohort study found higher dietary magnesium intake was linked to fewer externalizing behaviour problems in adolescents

Following 684 adolescents in the Western Australian Pregnancy Cohort (Raine) Study at ages 14 and 17, higher dietary magnesium intake was significantly associated with reduced externalizing behaviours after adjusting for sex, physical activity, family income and other factors, with a similar but non-significant trend for zinc intake.
