DIM & Estrogen Metabolism

DIM shifted estrogen metabolites in postmenopausal women

A study of 1,458 postmenopausal women using a transdermal estradiol patch found that the 108 women concurrently taking a DIM supplement had significantly altered urinary estrogen profiles, including changes in estrone, estriol, 2-OHE1, 2-OHE2, 4-OHE2, 16-OHE1 and the 2-OHE1/16-OHE1 ratio.

DIM & Estrogen Metabolism

Large cohort study of DIM and the urinary estrogen profile

Comparing 909 women reporting DIM use with 18,385 who did not, researchers found significant differences in nearly every urinary estrogen and estrogen metabolite, with the 2-hydroxyestrone:16-hydroxyestrone ratio increasing significantly with DIM use.

Calcium D-Glucarate & Detox Pathway

A natural beta-glucuronidase inhibitor

A clinical review describes calcium-D-glucarate as inhibiting beta-glucuronidase, an enzyme involved in Phase II liver detoxification; elevated beta-glucuronidase activity has been associated with increased risk of hormone-dependent cancers, and oral calcium-D-glucarate has been studied for regulating estrogen metabolism.

Calcium D-Glucarate & Detox Pathway

Four pathways behind D-glucaric acid's liver-detox role

A computational systems-biology analysis modeled how D-glucaric acid, the metabolite of calcium D-glucarate, supports liver detoxification through four molecular pathways: reduced reactive-oxygen production, reduced glucuronide deconjugation, lower hepatocyte apoptosis, and inhibition of beta-glucuronidase.

Broccoli & Cruciferous Compounds

Reviewing cruciferous compounds and breast cancer risk

An Annual Review of Nutrition analysis examined the evidence through 2024 for sulforaphane and indole-3-carbinol/DIM on breast cancer risk and outcomes, and discussed how individual variation in gut microbiome and glucosinolate metabolism may modify the benefits of cruciferous vegetable intake.

Broccoli & Cruciferous Compounds

How broccoli-derived indoles are hypothesized to shift estrogen metabolism

A review of indoles derived from glucobrassicin, found in broccoli and other crucifers, describes DIM's most well-characterized mechanism, modulation of aryl hydrocarbon receptor signaling, and notes that altered cytochrome P450-dependent estrogen metabolism is hypothesized to be an important driver of its studied breast-health effects.

Broccoli & Cruciferous Compounds

Sulforaphane reduced mammary tumor formation in an estrogen-exposed animal model

In rats co-exposed to 17beta-estradiol over 56 weeks, sulforaphane, a broccoli-derived compound, significantly protected against mammary tumor formation, reduced DNA damage, and lowered circulating free fatty acids and triglycerides.

Liver Support & Choline

Plasma choline tracks dietary intake and liver-fat response

In a controlled crossover feeding study, plasma choline and betaine concentrations reliably discriminated adequate from low dietary choline intake, and liver ultrasound elastography detected increased liver fat in a subset of participants on the lowest-choline diet arm.

Liver Support & Choline

Choline bitartrate supplementation improved liver fat in a clinical study

In patients with elevated homocysteine on standard therapy, 8 weeks of choline bitartrate supplementation (1000 mg/day) significantly raised plasma choline, lowered homocysteine, and was associated with regression of hepatic steatosis (fatty liver) in three of four affected patients.

Safety & Tolerability

DIM is well tolerated across a range of oral doses

Why the 2-hydroxylation pathway is considered the favorable one

Research on cytochrome P450 enzymes (CYP1A1, CYP1A2, CYP1B1) shows they convert estradiol into either 2-hydroxyestradiol, not associated with increased breast cancer risk, or 4-hydroxyestradiol, a pathway with genotoxic potential - the biochemical basis for why supporting the 2-hydroxylation route is a studied area of interest.
