Active Folate & Neural Tube Defects

The landmark trial: folic acid and neural tube defect prevention

A randomized, double-blind trial in women at high risk of a neural-tube-defect pregnancy found folic acid supplementation reduced recurrence by 72% compared with the groups that did not receive it.

Human RCT
MRC Vitamin Study Research Group, The Lancet (1991)
Active Folate & Neural Tube Defects

Periconceptional folic acid also linked to fewer congenital heart defects

A review of Hungarian intervention trials found periconceptional folic-acid-containing multivitamin supplementation prevented the majority of neural tube defects (about 90%) as well as a meaningful proportion of congenital heart defects (about 40%).

Review
Czeizel et al., Nutrients (2013)
Active Folate & Neural Tube Defects

Folate vs. folic acid: why the active form matters

A narrative review distinguishes dietary folate from synthetic folic acid and evaluates the evidence behind fortification and supplementation strategies for neural tube defect prevention, supporting a personalized approach to folate form and dose.

Review
Westmark, Nutrients (2026)
Gentle Iron & Maternal Blood Health

Daily iron supplementation reduces anemia risk in pregnancy

A Cochrane overview of systematic reviews found daily iron supplementation, with or without folic acid, increased hemoglobin levels and reduced the risk of anemia and iron-deficiency anemia in pregnant women, though intermittent regimens caused fewer side effects.

Systematic review (Cochrane)
Pena-Rosas et al., Cochrane Database of Systematic Reviews (2015)
Gentle Iron & Maternal Blood Health

Third-trimester anemia linked to lower Apgar scores and neonatal hospitalization

A retrospective study of pregnant women found third-trimester anemia was significantly associated with a lower 5-minute Apgar score and a higher risk of neonatal hospitalization, with about 80% of iron-deficiency anemia cases responding to supplementation.

Retrospective cohort study
Chang et al., Taiwanese Journal of Obstetrics & Gynecology (2026)
Gentle Iron & Maternal Blood Health

An honest null result: maternal anemia and early neonatal neurodevelopment

A systematic review and meta-analysis of 6 studies found no significant correlation between maternal anemia and neonatal neurodevelopmental outcomes assessed shortly after birth, noting that longer-term effects remain uncertain and warrant further research.

Meta-analysis
He & Liu, Asian Biomedicine (2026)
Choline & Fetal Brain Development

Systematic review: choline in pregnancy and child neurodevelopment

A systematic review of 4 randomized trials and 5 observational studies found most neurodevelopmental outcomes did not clearly support higher prenatal choline improving child brain development, though some individual trial endpoints favored supplementation - current evidence remains insufficient to confirm or refute a benefit.

Systematic review
Gould et al., Nutrients (2025)
Choline & Fetal Brain Development

RCT: prenatal choline supplementation and sustained attention at age 7

A controlled feeding trial randomized pregnant women to 480mg or 930mg of choline daily in the third trimester; children in the higher-choline group showed superior sustained attention on a signal-detection task at 7 years of age.

Human RCT (7-year follow-up)
Bahnfleth et al., FASEB Journal (2022)
Choline & Fetal Brain Development

Choline's expanding role in neurodevelopment and metabolic health

A research perspective describes converging evidence that maternal choline intake supports neurogenesis, cognition and visual system development, and may buffer the fetal brain against environmental and psychosocial stressors.

Review
Paules et al., Nutrients (2025)
Calcium, Vitamin D & Bone Health

Cochrane review: calcium supplementation and pre-eclampsia

An updated Cochrane review of 10 randomized trials (over 37,000 women) found calcium supplementation resulted in little to no difference in pre-eclampsia risk in sensitivity analyses restricted to larger, high-certainty trials.

Systematic review (Cochrane)
Cluver et al., Cochrane Database of Systematic Reviews (2025)
Calcium, Vitamin D & Bone Health

Maternal vitamin D status shapes neonatal and childhood outcomes

A narrative review describes how maternal vitamin D deficiency during pregnancy has been associated with adverse neonatal outcomes including low birth weight and poor skeletal growth, and with effects on neural and musculoskeletal development in the offspring.

Review
Vasdeki et al., Hormones (Athens) (2023)
Calcium, Vitamin D & Bone Health

Vitamin D deficiency remains a global issue for infant bone health

A narrative review notes vitamin D is indispensable for skeletal growth and bone health, with maternal and fetal vitamin D status affecting a substantial proportion of infants worldwide depending on geography and other risk factors.

Review
Stoica & Marginean, Nutrients (2023)
Iodine & Thyroid/Neurodevelopment

A century of iodine deficiency prevention

A historical review describes how iodine deficiency causes a spectrum of disease beyond goiter, including cretinism, intellectual impairment and adverse obstetric outcomes, and how salt iodization dramatically reduced global iodine deficiency disorders.

Review
Pearce & Zimmermann, Thyroid (2023)
Iodine & Thyroid/Neurodevelopment

Maternal iodine supplementation: mixed evidence on neurodevelopment

A review of maternal iodine supplementation trials in areas of mild-to-moderate iodine deficiency found no clear benefit on infant neurocognitive outcomes across meta-analyses, while noting women with restrictive diets may still be at risk for inadequate iodine status.

Review
Nguyen, Current Opinion in Endocrinology, Diabetes and Obesity (2023)
Iodine & Thyroid/Neurodevelopment

An honest null result: mild iodine insufficiency and child IQ at age 5

A secondary analysis of two randomized trials found maternal urinary iodine concentrations of 150mcg/L or less were not associated with lower full-scale IQ scores in children at 5 years of age among women with subclinical thyroid disease.

RCT (secondary analysis)
Casey et al., American Journal of Perinatology (2024)
B-Complex, B6 Balance & Pregnancy Wellness

Network meta-analysis: interventions for nausea and vomiting in pregnancy

A systematic review and network meta-analysis of 24 randomized trials found vitamin B6, alone or combined with ginger, acupressure or acupuncture, ranked among the more effective interventions for reducing symptoms of nausea and vomiting in pregnancy.

Network meta-analysis
Frivaldszky et al., Nutrients (2026)
B-Complex, B6 Balance & Pregnancy Wellness

RCT: doxylamine-pyridoxine for nausea and vomiting in pregnancy

A multicenter, randomized, placebo-controlled trial in 352 women with moderate-to-severe nausea and vomiting of pregnancy found doxylamine-pyridoxine produced a larger reduction in symptom scores than placebo, though the trial also flagged a signal for small-for-gestational-age birth warranting further study.

Human RCT
Wu et al., Annals of Internal Medicine (2023)
B-Complex, B6 Balance & Pregnancy Wellness

Vitamin B6 toxicity can occur even at modest multivitamin doses

A case report describes peripheral neuropathy in a patient whose only vitamin B6 source was a daily multivitamin providing 6mg - below the newly established tolerable upper intake level - underscoring why even standard multivitamin doses of B6 warrant a sensory-neuropathy caution.

Case report
Paluszny & Qiu, Cureus (2023)
Zinc, Antioxidants & Fetal Growth

Meta-analysis: zinc supplementation improves pregnancy and infant outcomes

A systematic review and meta-analysis of 77 randomized controlled trials found zinc supplementation in healthy pregnant women was associated with lower fetal growth-restriction rates, longer birth length, bigger head circumference and higher 1-minute Apgar scores.

Meta-analysis
Diao et al., Journal of Evidence-Based Medicine (2025)
Zinc, Antioxidants & Fetal Growth

Micronutrient supplementation and reducing low birth weight

A systematic review and meta-analysis of 43 studies found supplementation with vitamin D, zinc, or iron with folic acid each played a positive role in reducing the risk of low birth weight and small-for-gestational-age infants, with iron-folic acid showing the strongest and most consistent effect.

Meta-analysis
Sherzay et al., BMC Pregnancy and Childbirth (2026)
Zinc, Antioxidants & Fetal Growth

Antioxidant vitamin C and E supplementation shows mixed metabolic effects

A secondary analysis of a randomized trial of prenatal vitamin C and E supplementation found the effect on maternal and cord blood adiponectin, a metabolic hormone, depended on each woman's baseline adiponectin level - illustrating that antioxidant supplementation effects can be individualized rather than uniform.

Human RCT (secondary analysis)
Pippen et al., American Journal of Perinatology (2025)
Whole-Formula Safety & Periconceptional Use

Folic acid-containing multivitamins linked to fewer severe congenital heart defects

A national population-based case-control study found mothers who took high-dose folic acid during the critical period of heart development had significantly fewer children with ventricular septal defect, tetralogy of Fallot, d-transposition of the great arteries, and atrial septal defect.

Case-control study
Czeizel et al., European Journal of Obstetrics, Gynecology, and Reproductive Biology (2015)
Whole-Formula Safety & Periconceptional Use

Maternal B12, vitamin D and folate status together shape child neurodevelopment

A systematic review of 22 studies found adequate maternal vitamin B12 status was consistently associated with improved language, memory and cognitive performance, while periconceptional folic acid supplementation showed the most consistent favorable associations with cognitive and language development.

Systematic review
Franco et al., Frontiers in Neuroscience (2026)
