Meta-analysis: melatonin improves sleep onset latency in delayed sleep phase disorder
A systematic review and meta-analysis of 34 randomized controlled trials found that melatonin significantly improved sleep onset latency, measured by diary, and total sleep time, measured by polysomnography, in adults with delayed sleep phase disorder.
Meta-AnalysisMeta-analysis finds melatonin's most convincing evidence is for reducing sleep onset latency
A meta-analysis of randomized, controlled studies found the most convincing evidence for exogenous melatonin was in reducing sleep onset latency in primary insomnia and delayed sleep phase syndrome, compared with placebo.
Meta-AnalysisReview of 19 RCTs: melatonin improved sleep latency 22-60 minutes in children with DSPD
A review of 19 randomized controlled trials in 841 children and adolescents with delayed sleep-wake phase disorder found that melatonin treatment, usually over 4 weeks, consistently improved sleep latency by 22 to 60 minutes without serious adverse effects.
ReviewCochrane review: melatonin is remarkably effective at preventing and reducing jet lag
A Cochrane systematic review of 10 randomized trials in air travellers found that melatonin, taken close to the target bedtime, decreased jet lag from flights crossing five or more time zones in 9 of 10 trials, with daily doses between 0.5 and 5 mg similarly effective and a low incidence of other side effects.
Cochrane ReviewSystematic review: melatonin supplementation helps realign circadian rhythms disrupted by travel
A systematic review of 23 studies on circadian disruption from trans-meridian travel and shift work found that interventions including melatonin supplementation, personalized light exposure and chrono-modulation significantly improved sleep quality and reduced jet lag symptoms.
Systematic ReviewReview links night-shift work to suppressed melatonin and disrupted sleep quality
A systematic review of 14 studies in healthcare professionals found that night-shift work was consistently associated with suppressed melatonin secretion, circadian misalignment and poorer sleep quality, and noted that melatonin supplementation to improve sleep quality may help mitigate this occupational circadian disruption.
Systematic ReviewMeta-analysis finds a good overall safety profile for higher-dose melatonin
A systematic review and meta-analysis of 79 randomized trials (3,861 adult participants) found that higher-dose melatonin (10 mg or more) did not increase serious adverse events or study withdrawals compared with placebo, though it modestly increased milder effects such as drowsiness, headache and dizziness.
Meta-AnalysisMeta-analysis: perioperative melatonin reduced postoperative delirium without severe adverse events
A systematic review and meta-analysis of 16 randomized controlled trials (1,981 patients) found that perioperative melatonin suppressed postoperative delirium, particularly at higher doses, without causing severe adverse events.
Meta-AnalysisReview: melatonin has strong evidence and fewer drug interactions among sleep medications used in hospital
A review of medications given for sleep in hospitalized patients found melatonin was the most commonly used agent and had the strongest evidence for use, with a comparatively low likelihood of adverse effects and drug-drug interactions relative to benzodiazepines and other sedative-hypnotics.
ReviewCombined GABA and L-theanine improved sleep quality over 4 weeks
An exploratory study in adults with sleep problems found that combined daily intake of GABA (700 mg) and L-theanine (200 mg) for 4 weeks significantly improved total Pittsburgh Sleep Quality Index scores and wearable-tracked sleep recovery scores.
Human TrialA 50mg GABA and botanical combination improved sleep quality in menopausal women
A randomized, double-blind, placebo-controlled study in 80 women found that a nutraceutical combining 50 mg of GABA with a botanical extract significantly improved sleep quality, stress and mood-related symptoms compared with placebo over 5 weeks of daily use.
Human RCTReview: GABA is the major inhibitory neurotransmitter and supports relaxation and insomnia prevention
A review of GABA receptor biology describes gamma-aminobutyric acid as the major inhibitory neurotransmitter in the mammalian nervous system, noting its roles in promoting neuronal relaxation and helping to prevent insomnia.
ReviewA single 200mg dose of L-theanine reduced cortisol and increased relaxation-linked brain activity under stress
A randomized, triple-blind, placebo-controlled crossover study found that a single 200 mg dose of L-theanine significantly increased frontal-region alpha brain wave activity and reduced salivary cortisol after an acute stress challenge, compared with placebo.
Human RCTAn L-theanine drink reduced subjective stress response to a cognitive stressor
A randomized, double-blind, placebo-controlled crossover trial in 34 healthy adults found that an L-theanine-based drink significantly reduced subjective stress response and salivary cortisol following a cognitive stress test, compared with placebo.
Human RCTL-theanine showed relaxing effects under resting conditions in a comparison with a standard anxiolytic
A double-blind, placebo-controlled study comparing 200 mg of L-theanine with alprazolam found some evidence of relaxing effects for L-theanine on a tranquil-troubled mood measure under resting conditions, though neither substance showed anxiolytic effects during an experimentally induced anxiety state.
Human TrialLemon balm supplementation improved sleep quality and mood over 3 weeks
A clinical study in 32 adults with poor sleep quality and emotional distress found that daily supplementation with a lemon balm (Melissa officinalis) extract for 3 weeks significantly improved Pittsburgh Sleep Quality Index scores, with the greater benefit seen at the higher (400 mg/day) dose, alongside reductions in depression, anxiety and stress scores.
Human TrialA standardized lemon balm extract reduced insomnia severity in a placebo-controlled crossover study
A prospective, double-blind, placebo-controlled crossover study found that a standardized lemon balm (Melissa officinalis) extract significantly reduced Insomnia Severity Index scores and increased slow-wave sleep duration compared with placebo, with 87% of treated participants reporting improved sleep quality.
Human RCTIntegrative review: lemon balm shows the most consistent clinical evidence for anxiety, stress and sleep
An integrative review of lemon balm (Melissa officinalis) phytochemistry and clinical research found that its most consistent clinical signals, across controlled trials, were for anxiety/stress and sleep-related outcomes, supporting continued use of standardized preparations.
ReviewSystematic review: passionflower reduced anxiety across nine clinical trials
A systematic review of nine randomized clinical trials found that passionflower (Passiflora incarnata) preparations reduced anxiety levels in most studies, with the literature suggesting it may also help with insomnia and depression-related symptoms, and no adverse effects on memory or psychometric function were observed.
Systematic ReviewValerian root extract improved sleep quality after cardiac surgery
A triple-blind randomized controlled trial in 72 patients found that 530 mg of valerian root extract taken nightly for 30 nights after coronary artery bypass graft surgery significantly improved total sleep quality, sleep latency, sleep duration and sleep efficiency compared with placebo, with no safety concerns identified.
Human RCTReview: valerian shows some evidence for insomnia and a well-established safety profile
A review of clinical practice guidelines, systematic reviews and randomized trials found some evidence for valerian's effectiveness in treating insomnia, while evidence for anxiety disorders was insufficient; valerian root was found to be well tolerated and safe, with infrequent and benign side effects.
ReviewSkullcap extract showed anxiolytic effects linked to GABA-A receptor binding compounds
A phytochemical and behavioral study found that Scutellaria lateriflora (American skullcap) extracts produced anxiolytic-like effects in animal models, and identified the flavonoids baicalin and baicalein - compounds known to bind the benzodiazepine site of the GABA-A receptor - alongside GABA itself, as likely contributors to this activity.
ReviewReview of herbal anxiolytics and sedatives highlights both benefits and drug-interaction cautions
A narrative review of clinical data on plant-based supplements used for anxiety, depression and insomnia found meaningful improvements in mood, stress and sleep quality across studies, while cautioning that herb-drug interactions (via cytochrome P450 pathways), excessive sedation and product quality are real safety considerations requiring clinical awareness.
ReviewScoping review: valerian and melatonin have the strongest evidence among over-the-counter sleep aids
A scoping review of 51 randomized controlled trials of over-the-counter products for insomnia found that, among the many products studied, valerian and melatonin had the most substantial evidence for effectiveness and safety, with promising results also seen for combinations such as valerian with lemon balm, and valerian with passionflower.
