Curcumin & Joint Health

A 2026 Australian RCT found a curcumin-containing combination no better than placebo for knee pain

The 12-week, placebo-controlled ATLAS trial gave 84 adults with knee osteoarthritis a daily combination including 500 mg curcumin (plus Boswellia, pine bark, MSM and piperine) or placebo; both groups improved similarly on a pain scale, with no significant between-group difference - an honest null result for this particular multi-ingredient combination.

Human RCT
Yuan et al., Osteoarthritis and Cartilage (2026)
Curcumin & Joint Health

A network meta-analysis of 39 trials (4,599 patients) ranked curcumin among the more effective knee-OA supplements

Comparing seven nutritional supplements for knee osteoarthritis - eggshell membrane, vitamin D, Boswellia, curcumin, ginger, krill oil and collagen - curcumin and krill oil showed the most promise for improving function, though Boswellia ranked most effective overall for pain and stiffness; no supplement increased adverse events versus placebo.

Network meta-analysis
Zhang et al., Nutrients (2025)
Curcumin & Joint Health

An 8-week RCT found a standardized curcumin extract reduced knee pain in 101 adults

Twice-daily 500 mg doses of a standardized curcumin extract significantly reduced knee-pain scores and numeric pain ratings versus placebo, and 37% of curcumin users cut their pain medication compared with 13% on placebo.

Anti-Inflammatory Mechanisms

Bisdemethoxycurcumin, a natural curcumin relative, eased arthritis in rats more effectively than curcumin itself

In an adjuvant-induced arthritis model, this minor turmeric curcuminoid reduced paw swelling, arthritic index and pro-inflammatory cytokines more effectively than curcumin, working by blocking IkBa degradation, lowering the NF-kB p65 ratio, and inhibiting macrophage migration.

Animal study
Sun et al., Chemico-Biological Interactions (2023)
Anti-Inflammatory Mechanisms

Curcumin blocked NF-kB and NLRP3 inflammasome activation in human dental pulp stem cells

In lipopolysaccharide-stimulated human cells, curcumin dose-dependently reduced IL-1beta and NLRP3 expression by inhibiting phosphorylation of NF-kB p65, a core inflammatory signaling step relevant to curcumin's broader anti-inflammatory research base.

In vitro study
Lan et al., Clinical Oral Investigations (2023)
Anti-Inflammatory Mechanisms

Intranasal curcumin eased chronic asthma in mice via NF-kB and MAPK pathways

In an ovalbumin-induced chronic-asthma model, curcumin reduced oxidative markers in lung fluid and blocked phosphorylation of JNK, ERK1/2 and p38, thereby lowering NF-kB activation and COX-2 expression in lung tissue.

Animal study
Chauhan et al., Phytomedicine (2018)
Antioxidant & Oxidative Stress

A dose-response meta-analysis of 28 RCTs found curcumin lowered inflammatory and oxidative markers

Pooling 31 effect sizes, curcumin/turmeric supplementation significantly reduced CRP, TNF-alpha, IL-6 and malondialdehyde while raising glutathione and total antioxidant capacity in people with prediabetes or type 2 diabetes - benefits were greatest with unformulated curcumin at 1 g/day or more, though overall certainty was rated low.

Meta-analysis
Bahari et al., Inflammopharmacology (2025)
Antioxidant & Oxidative Stress

A 2026 review found curcumin may help exercise recovery, though evidence is mixed

Across 15 trials in active adults and athletes, curcumin favorably affected oxidative-stress markers in most studies (6 of 7), with more inconsistent results for muscle damage, inflammation and performance outcomes - the certainty of evidence was rated low to very low.

Systematic review
Lloret-Gil, Victoria-Montesinos & Martinez-Noguera, Nutrients (2026)
Antioxidant & Oxidative Stress

A review found turmeric/curcumin supported antioxidant status in dialysis patients

Across 21 studies in hemodialysis patients and those on immunosuppressive drugs, turmeric and curcumin showed antioxidant, anti-inflammatory and nephroprotective benefits - though the reviewers also flagged that a food-drug interaction between high-dose turmeric and immunosuppressants caused nephrotoxicity in one instance, underscoring the importance of following labeled doses.

Review
Tran & Tran, Anti-Cancer Agents in Medicinal Chemistry (2020)
Bioavailability & Water-Solubility Technology

A pharmaceutics review outlines nanocarrier approaches developed to overcome curcumin's low oral bioavailability

Curcumin's benefits are limited by poor water solubility and rapid metabolism; this review discusses polymeric nanomicelle delivery systems designed specifically to improve curcumin's solubility and bioavailability, illustrating why delivery technology matters as much as the ingredient itself.

Review
Farhoudi et al., International Journal of Pharmaceutics (2022)
Bioavailability & Water-Solubility Technology

A formulation study found engineering curcumin into a water-soluble nanofiber increased its solubility up to 38-fold

Comparing two curcumin nanoformulation strategies in lab testing, a water-soluble polymer-based nanofiber format increased curcumin's solubility 16 to 38 times versus untreated curcumin - a concrete illustration of how re-engineering curcumin's physical form can address its naturally poor water solubility.

Laboratory study
Rawas-Qalaji et al., Colloids and Surfaces B: Biointerfaces (2024)
Cardiovascular & Lipid Health

A meta-analysis of 27 RCTs found curcumin modestly improved lipid profiles in diabetes/prediabetes

Pooling 31 trial arms, curcumin/turmeric supplementation significantly lowered triglycerides, total cholesterol and LDL-C while raising HDL-C in adults with prediabetes or type 2 diabetes - benefits were larger in diabetic patients and those with higher baseline lipids, though certainty of evidence was rated low.

Meta-analysis
Bahari et al., Lipids (2026)
Cardiovascular & Lipid Health

Adding curcumin to standard diabetes therapy improved lipids and inflammation in a 60-patient RCT

In type 2 diabetes patients on glimepiride, adding 1,100 mg curcumin daily for three months significantly reduced total cholesterol, triglycerides, LDL-C and hs-CRP compared to placebo, performing similarly to (though slightly less strongly than) a comparator fibrate drug.

Human RCT
Nada et al., BMC Pharmacology & Toxicology (2025)
Cardiovascular & Lipid Health

A 227-patient, 12-month RCT found curcumin reduced atherogenic risk markers in obese type 2 diabetics

Curcumin supplementation significantly lowered pulse wave velocity (a marker of arterial stiffness), LDL-C, small dense LDL-C, hs-CRP and inflammatory cytokines compared with placebo over 12 months in obese adults with type 2 diabetes.

Human RCT
Yaikwawong et al., Nutrients (2024)
Metabolic & Glycemic Health

A triple-blind RCT found curcumin plus piperine did not significantly change glucose control in prediabetes

Over three months, 500 mg curcumin plus 5 mg piperine daily lowered CRP but produced no significant change in fasting glucose or HbA1c versus placebo in prediabetic adults - an honest result showing this combination's glycemic effect was not established in this trial.

Human RCT
Ghazaee et al., Avicenna Journal of Phytomedicine (2026)
Metabolic & Glycemic Health

A meta-analysis of 14 RCTs (1,277 subjects) found curcumin improved glucose metabolism in cardiovascular-risk patients

Curcumin supplementation significantly reduced fasting blood glucose, HbA1c and insulin resistance (HOMA-IR) in people with cardiovascular risk factors, with the strongest effects at doses of 300 mg/day or more taken for 12 weeks or longer; curcumin was well tolerated with no serious adverse events.

Meta-analysis
Huang et al., Journal of Human Nutrition and Dietetics (2019)
