What the research says about the B-vitamins and cofactors in Advanced B Complex Ultra
Peer-reviewed studies and reviews on the individual B-vitamins and cofactors in this formula - PQQ, the core B-vitamins, choline and myo-inositol - shared for education, not as claims about this finished product. Advanced B Complex Ultra's claims are the statements authorized under its Natural Health Product licence, NPN 80068201.
Improved memory, attention and cognitive flexibility
A 12-week randomized, double-blind, placebo-controlled trial of PQQ disodium salt (about 21.5 mg/day) in healthy adults aged 40 to under 80 found significant improvements versus placebo across composite memory, verbal memory, reaction time, complex attention, cognitive flexibility, executive function and motor speed.
Human RCTAge-dependent pattern of cognitive benefit
A 12-week randomized, placebo-controlled trial of PQQ disodium salt (20 mg/day) in adults aged 20 to 65 found improved composite and verbal memory overall, with younger adults (20-40) showing gains in cognitive flexibility and processing speed by 8 weeks and older adults (41-65) showing memory gains by 12 weeks.
Human RCTRaised BDNF and brain oxygenation in mild cognitive impairment
A 6-week randomized, placebo-controlled trial of a dihydrogen-PQQ combination in elderly adults with mild cognitive impairment found a significant rise in serum BDNF, improvement on the orientation domain of a standard Alzheimer's cognitive scale, and increased cerebral oxygen saturation versus placebo.
Human RCTA mitochondrial-biogenesis mechanism
A 2026 review describes PQQ as a mitochondrial and redox regulator that enhances mitochondrial biogenesis and bioenergetic capacity via the AMPK and SIRT1/PGC-1-alpha signaling pathways, positioning it among nutrients studied for healthy aging.
ReviewGreater stamina, concentration and alertness at work
In a randomized, placebo-controlled trial of 198 working men, a B-vitamin complex combined with vitamin C and minerals was associated with higher self-rated physical stamina, concentration and mental stamina during the working day, and greater alertness, versus placebo over 28 days.
Human RCTThiamine and energy intake in older adults
A double-blind, placebo-controlled trial in elderly women with marginal thiamine deficiency found that 10 mg/day of thiamine for 6 weeks significantly increased appetite, energy intake, body weight and general well-being, and reduced fatigue, versus baseline and placebo.
Human RCTA niacin-related route to raising cellular NAD+
A randomized, placebo-controlled crossover pilot trial of a nicotinamide (vitamin B3) and D-ribose combination in healthy middle-aged adults found a significant rise in the blood NAD+ metabolome alongside less self-reported fatigue and improved mental concentration and motivation.
Human RCTBioactive folate forms for vascular and homocysteine health
A 2025 review of vitamins and vascular health describes how the bioactive folate forms L-5-methyltetrahydrofolate (L-methylfolate) and levoleucovorin can support one-carbon metabolism and the conversion of homocysteine to methionine when conversion from synthetic folic acid is a limiting step.
ReviewThe body already relies on phosphorylated, coenzyme B-vitamin forms
An analysis of human milk across lactation stages measured multiple chemical forms of thiamine, riboflavin and folate side by side - including phosphorylated/coenzyme forms such as thiamine pyrophosphate, FAD and 5-methyltetrahydrofolate - confirming the body naturally circulates these active forms, not only the parent vitamins.
Human studyWhy the active B12 form matters when methylation is impaired
Updated clinical guidelines for inherited disorders of vitamin B12/folate metabolism identify high-dose hydroxocobalamin - an active, ready-to-use form of B12 - as first-line therapy that may improve neurocognitive outcomes; specific to these rare inherited conditions, this illustrates why the chemical form of a B-vitamin can matter for how the body uses it.
Clinical guidelineAn established role in brain development, a plausible role across life
A narrative review describes choline as a critical nutrient for early neurodevelopment with a plausible, though not yet fully established in humans, role in lifelong cognitive function, acting partly through epigenetic programming via DNA and histone methylation; typical intakes often fall short of recommendations.
ReviewModerate choline intake linked to lower dementia risk
A prospective cohort study of over 5,300 older adults plus a dose-response meta-analysis of five cohorts (over 137,000 participants) found an L-shaped association between dietary choline intake and dementia risk, with moderate intake associated with lower risk than the lowest intake.
Human cohort + meta-analysisAn inositol-containing supplement and gaming-related cognitive performance
A double-blind, placebo-controlled trial of an inositol-enhanced arginine-silicate supplement in healthy video gamers found improved accuracy on attention-switching tasks, reduced fatigue and anger, and increased self-reported energy after a week of use, with minimal adverse events.
Human RCTAn honest look at the limits: no effect on mood in a large prenatal trial
The NiPPeR randomized trial of a myo-inositol, probiotic and micronutrient formulation taken from preconception through pregnancy found no significant difference in mood or anxiety scores versus a standard micronutrient supplement, though it did find a modest improvement in a general mental-health-functioning score.
Human RCTB12 supplementation reliably corrects deficiency
A Cochrane systematic review of 16 randomized trials (4,083 children) found vitamin B12 supplementation reduced B12 deficiency with high-certainty evidence and probably increased B12 concentrations, though it found little to no difference in growth or cognitive-test scores in this population.
Meta-analysisFolate and B12's role in nerve function and blood cell formation
A review of nutrition and frailty in older adults describes vitamin B12 as essential for nerve function and red blood cell formation, and folate as crucial for DNA synthesis and cell division, while noting evidence on supplementation specifically preventing frailty remains mixed.
ReviewHow the body moves folate and B12 from mother to newborn
A study of maternal weight status and neonatal nutrient status found lower red-blood-cell folate in mothers with overweight/obesity, along with reduced newborn folate forms, total B12 and holotranscobalamin, and identified altered placental folate/B12 transporter expression as a likely mechanism.
Human studyNiacin improved LDL, total cholesterol and Lp(a) in a clinical trial
A randomized controlled trial of niacin (500 mg/day) added to standard care in hemodialysis patients over 3 months found significantly greater reductions in LDL cholesterol (31.9% vs 7.9%), total cholesterol (13.3% vs 3.7%) and lipoprotein(a) (11.4%) compared with standard care alone.
Human RCTWhy niacin causes flushing - and its lipid benefit
A 2024 mechanistic review explains that niacin-induced skin flushing occurs when niacin activates the HCA2 (GPR109A) receptor, triggering prostaglandin release and cutaneous vasodilation, and separately describes niacin's established benefit of improving lipid profiles when used alongside statin therapy.
ReviewNiacin among the therapies shown to raise HDL cholesterol
A 2026 review of lipid-lowering therapies notes niacin is among the treatments demonstrated to substantially raise HDL cholesterol alongside lowering triglycerides, historically used for correcting dyslipidemia.
ReviewB12 is generally well tolerated, with limits on who benefits
A 2026 review examining whether more B12 is always better found supplementation is generally well tolerated with only rare reports of acneiform or hypersensitivity reactions, while also finding no consistent benefit for fatigue, mood, cognition or cardiovascular outcomes in people without a biochemical deficiency.
ReviewFolate's safety profile across 1,367 people
A systematic review of folate supplementation for peripheral neuropathy pooling multiple studies reported only one adverse event across 1,367 participants, alongside reductions in homocysteine and inflammatory markers - an excellent safety and tolerability profile for the doses studied.
Systematic reviewA vitamin B-complex was well tolerated in a placebo-controlled trial
A randomized, double-blind, placebo-controlled trial of a vitamin B-complex combined with a non-hormonal supplement in perimenopausal women reported the combination was well tolerated with only mild, transient side effects, alongside improvements in mood, anxiety and sleep-related scores.
Human RCTWhy these vitamins are biochemically essential cofactors
A 2026 review of inherited cofactor disorders maps how vitamin B6 (as pyridoxal-5-phosphate), niacin (as NAD) and pantothenate (as coenzyme A) each function as essential metabolic cofactors, using rare genetic defects in their pathways to illustrate the wide-ranging biochemical roles these nutrients normally play.
ReviewRiboflavin's coenzyme (FAD) role across metabolism
A systematic review of riboflavin therapy across 33 inherited metabolic diseases found clear effectiveness in several riboflavin-responsive conditions, underscoring how central the riboflavin-derived coenzyme FAD is to numerous cellular metabolic processes.
Systematic reviewThiamine's coenzyme form fuels a key metabolic pathway
A methodological review of transketolase-activity testing explains that thiamine diphosphate - the active coenzyme form of thiamine (vitamin B1) - is required by transketolase, a rate-limiting enzyme of the pentose-phosphate pathway used to assess functional thiamine status.
