What the research says about this 5-active formula
Educational summaries of published research on palmitoylethanolamide (PEA), Univestin, Boswellia serrata, quercetin, magnesium glycinate, and joint/pain-support formulas generally - not product-specific claims. AOR P.E.A.K Pain Relief's own claim is the authorized use under NPN 80094703: magnesium for bone/muscle health, quercetin as an antioxidant/capillary protectant, PEA studied as an anti-inflammatory for chronic pain, and Univestin for joint discomfort, stiffness and mobility in osteoarthritis.
A meta-analysis of 774 patients found PEA eased chronic pain
A systematic review and meta-analysis of 11 double-blind randomized controlled trials (774 patients total) found palmitoylethanolamide (PEA) reduced pain intensity scores versus comparators, with several studies reporting added benefits for quality of life and functional status and no major side effects attributed to PEA in any study.
Meta-analysisAn 8-week trial found a PEA formulation eased neuropathic low back pain
In a double-blind, placebo-controlled trial of 120 adults with chronic neuropathic low back pain, a phospholipid-based PEA formulation significantly improved pain, functional disability, sleep quality and quality-of-life scores versus placebo over 8 weeks, and was associated with more frequent discontinuation of other analgesics.
Human RCTPEA is described as a well-tolerated lipid mediator supporting joint health
A review of palmitoylethanolamide (PEA) research describes it as an endocannabinoid-like lipid mediator with documented anti-inflammatory, analgesic and immunomodulatory effects across multiple targets, noting applications in pain modulation and joint health, and reporting it is well tolerated and devoid of side effects in animal and human studies.
ReviewA 1-week trial found the Univestin blend eased knee pain as well as naproxen
In a double-blind randomized trial, 79 adults with mild-to-moderate knee osteoarthritis took either 500 mg/day of UP446 - the standardized Scutellaria baicalensis and Acacia catechu blend behind Univestin - or 440 mg/day of naproxen for one week; the UP446 group showed a significant, time-dependent decrease in perceived pain and a significant increase in knee range of motion.
Human RCTThe Scutellaria-catechu extract blocks two inflammatory enzymes at once
Laboratory testing of a mixed extract of baicalin (from Scutellaria baicalensis) and catechin (from Acacia catechu) found it inhibited both cyclooxygenase (COX-1/COX-2) and 5-lipoxygenase enzyme activity, reduced prostaglandin E2 and leukotriene production in human cell lines, and decreased swelling in an arachidonic-acid-induced inflammation model.
Lab & animal studyA 12-week trial supported the safety of the same botanical blend in knee osteoarthritis
A randomized, double-blind, placebo-controlled trial of flavocoxid - a proprietary blend of Scutellaria baicalensis and Acacia catechu flavonoids - in 59 patients with knee osteoarthritis found no significant differences in adverse events, blood pressure or laboratory values compared with placebo over 12 weeks, supporting its safety profile.
Human RCTA standardized boswellia extract eased knee pain within 7 days
In a 90-day, double-blind, placebo-controlled trial, a Boswellia serrata extract enriched with boswellic acids (5-Loxin) produced significant improvements in pain and physical function as early as 7 days, alongside a reduction in the cartilage-degrading enzyme MMP-3 in joint synovial fluid, with safety parameters unchanged versus placebo.
Human RCTA pharmacology review maps how boswellia's resin may fight inflammation
A comprehensive review of Boswellia serrata gum resin extract across in vitro, preclinical, pharmacokinetic and clinical studies describes its long traditional use and potential as an alternative to NSAIDs for inflammatory conditions, while examining open questions about exactly which boswellic acids and enzyme targets drive its anti-inflammatory effects.
ReviewA refined boswellia extract showed better absorption and cartilage protection
Laboratory and animal testing comparing two standardized Boswellia serrata extracts found the newer composition (Aflapin) increased the bioavailability of its key boswellic acid by over 50% versus a standard 30%-boswellic-acid extract, and provided better protection of human cartilage cells from inflammatory breakdown.
Lab & animal studyQuercetin calmed inflammatory signaling in cartilage cells
Laboratory and animal research on knee osteoarthritis found quercetin reduced interleukin-1beta-induced inflammation and cartilage matrix breakdown in chondrocytes by downregulating a specific transcription factor (E2F2) and modulating downstream inflammatory signaling, supporting further study of quercetin as a joint-supportive compound.
Lab & animal studyA review of 31 human studies mapped what actually boosts quercetin absorption
A systematic review and meta-analysis of 31 human intervention studies found quercetin's bioavailability varies substantially by chemical form and formulation - glucoside forms absorbed far better than the plain aglycone, and taking quercetin with dietary fat or fiber roughly doubled absorption - informing how quercetin supplements are best formulated and taken.
Systematic reviewA crossover trial found magnesium glycinate absorbs via a distinct gut pathway
A double-blind, randomized crossover trial in patients with prior intestinal (ileal) resection - a population prone to poor magnesium absorption - found magnesium diglycinate (the glycinate chelate form) was absorbed significantly better than magnesium oxide in those with the most impaired absorption, with evidence it is taken up in part intact via a dipeptide transport pathway, and was better tolerated by all patients.
Human RCTA multi-ingredient formula combining PEA and quercetin protected cartilage in an OA model
In a rat osteoarthritis model, a multi-component formula containing olive-leaf bioflavonoids including quercetin, palmitoylethanolamide, hyaluronic acid and other actives significantly reduced joint degeneration markers and inflammatory signaling (the NLRP3-TXNIP-iNOS axis) compared with untreated controls, performing comparably to other tested nutraceuticals and ibuprofen on several measures.
Animal studyA curcumin-boswellia combination eased hand osteoarthritis pain over 3 months
In a 3-month, multicenter, placebo-controlled trial of 162 patients with hand osteoarthritis, a combination of patented Curcuma longa and Boswellia serrata extracts significantly reduced pain on a visual analog scale versus placebo, with comparable tolerability between groups.
Human RCTA 5-ingredient joint combination, including boswellia, did not outperform placebo
The 12-week ATLAS trial tested a combination of Boswellia serrata, pine bark extract, methylsulfonylmethane, curcumin and piperine against placebo in 84 adults with symptomatic knee osteoarthritis; both groups improved similarly, and the combination was not shown to be superior to placebo for pain - an honest reminder that not every plausible-sounding joint blend outperforms placebo in a rigorous trial.
Human RCTA 26-week safety study found no toxicity signal for the Univestin blend
A 26-week repeated-dose oral toxicity study of UP446 (the Scutellaria baicalensis/Acacia catechu blend marketed as Univestin) in beagle dogs at up to 1000 mg/kg/day found no mortality, and no significant differences in body weight, bloodwork, organ weights or pathology versus control, establishing a no-observed-adverse-effect level of 500 mg/kg/day.
Animal safety studyA meta-analysis found PEA favorably tolerated across diabetic nerve-pain trials
A systematic review and meta-analysis of 3 studies (188 patients) evaluating palmitoylethanolamide for diabetic neuropathic pain found consistent, significant reductions in pain scores alongside a favorable tolerability profile, with safety data indicating only mild, self-limited adverse events.
Meta-analysisA boswellic-acid combination showed a good safety profile against a glucosamine comparator
In a randomized trial of 120 participants with knee arthritis, a combination of methylsulfonylmethane and boswellic acids taken for 60 days showed pain and function improvements comparable to or better than glucosamine sulfate, with a favorable safety and tolerability profile monitored throughout the study.
Human RCTBoswellia ranked most effective among 7 joint-health supplements
A network meta-analysis of 39 randomized trials (4,599 patients) comparing 7 common nutritional supplements for knee osteoarthritis found Boswellia had the highest probability of being most effective for pain and stiffness, with curcumin, collagen, ginger and krill oil also showing benefit on some measures, and no supplement associated with increased adverse events versus placebo.
Network meta-analysisA review maps the plant compounds studied for osteoarthritis
A review of bioactive plant compounds for osteoarthritis - including Boswellia serrata, curcumin, ginger and resveratrol - describes their shared anti-inflammatory, antioxidant and tissue-protective mechanisms and the molecular pathways through which they may help modulate joint inflammation and pain.
ReviewA comparison of 21 joint supplements ranked several natural options highly
A network meta-analysis of 23 randomized trials (2,455 participants) comparing 21 different dietary supplements for osteoarthritis found methylsulfonylmethane, undenatured type II collagen and collagen ranked among the top options for improving WOMAC pain and function scores, though the authors noted the overall evidence quality was low and called for larger trials.
Network meta-analysisA review reframes joint supplements as adjuncts, not stand-alone cures
A narrative review examining commonly used oral nutraceuticals - including Boswellia, curcumin, collagen peptides and omega-3s - through a muscle-joint framework found symptomatic benefits were modest, heterogeneous and formulation-dependent, positioning them as optional, time-limited adjuncts that may support rehabilitation participation rather than stand-alone treatments.
ReviewA review maps where nutraceuticals fit in the broader osteoarthritis toolkit
A review of therapeutic advances in osteoarthritis management surveys pharmacologic treatments, nutraceuticals, biologics and exercise together, highlighting nutraceuticals as one part of a broader, multi-pronged strategy for mitigating disease progression and supporting rehabilitation.
ReviewOrganic magnesium compounds absorb differently depending on their chemical form
Animal research comparing organic-acid-bound magnesium compounds (citrate, malate) with amino-acid-bound compounds (magnesium glycinate, acetyl taurate) found meaningful differences in tissue magnesium levels by compound and dose, underscoring that the chemical form a magnesium supplement uses affects how much reaches the body's tissues.
